The two-year veteran was hospitalized after surgery at Phoenix's Maricopa County Medical Center, state Department of Public Safety spokesman Frank Valenzuela said.
Cardinals vice president Michael Bidwill said Anderson was in serious condition.
According to Valenzuela, Anderson steered his Ford Expedition toward an Interstate 10 off-ramp in Chandler about 2:30 a.m. He lost control, and the vehicle rolled down an embankment and came to rest on its roof.
Anderson and his brother, Billy, were thrown from the vehicle. Another brother, Dedric, remained inside. Valenzuela said Billy and Dedric had surgery for leg injuries.
Damien Anderson, 24, was Arizona's backup running back until the Cardinals signed Emmitt Smith last March.
Relegated to third-string, Anderson finished fifth on the team in rushing, gaining 68 yards in 18 carries.
